for a secondary control step. This control would normally be a function of changes in porphobilinogen deaminase activity, since uroporphyrinogen co-synthase activity is normally about 10 times that of porphobilinogen deaminase activity (Frydman & Feinstein, 1974). Thus, in situations when 6-aminolaevulinate synthase activity is derepressed, porphobilinogen deaminase activity may be limiting and may provide a secondary control mechanism. This mechanism does not come into effect in patients with variegate porphyria and hereditary coproporphyria for reasons which can only at present remain speculative. The present study does not exclude the possiblilty of important changes in uroporphyrinogen co-synthase activity or in the combination of interrelationships of the two enzymes active at this site in haem biosynthesis.
